个人简介

南方科技大学医学院副教授、博士生导师,2013年博士毕业于北京大学生命科学学院,随后分别在芝加哥大学和费城儿童医院/宾夕法尼亚大学进行博士后研究,于2022年6月加入南方科技大学医学院。主要研究造血干细胞(HSC)功能调控及相关血液病,如髓系白血病和骨髓衰竭综合征等。

近几年在HSC和血液病领域主要取得以下研究成果:1)揭示泛素连接酶HECTD1调控的核糖体装配在HSC再生过程中的功能,并为SDS亚型的骨髓衰竭综合征提供了基因治疗靶点(Cell Stem Cell, 2021);2)阐明致癌突变体FLT3-ITD的棕榈酰化修饰在急性髓系白血病中的功能(Blood, 2021);3)发现JAK2激酶在HSC和髓系白血病中的泛素化降解机制,并证明JAK2的抑制剂可以“老药新用”治疗CBL和LNK基因突变导致的多种白血病(Genes & Development, 2017)。另受Blood杂志社邀请以第一作者身份撰写评论(Commentary)文章1篇(Blood,2021)。

研究领域

造血干细胞是骨髓中一群数量极少、但具有自我增殖和分化功能的起始细胞,是体内几乎所有血细胞的来源。造血干细胞功能失控会导致多种血液病,对人类健康造成严重危害。本课题组主要研究细胞因子/癌基因信号通路和核糖体发生、装配调控的蛋白稳态在正常或癌变条件下对造血干细胞功能和疾病发生的影响。我们以转基因小鼠、原代病人细胞或永生化的造血细胞系为模型,重点使用小鼠骨髓移植、polysome profiling/RNA-Seq、蛋白相互作用复合体的纯化并结合质谱分析等多种生物化学、分子生物学和细胞生物学手段对以上问题进行研究,并在此基础上探索血液病治疗的新方法。

1)信号蛋白的翻译后修饰对正常和疾病条件下造血发生的影响。

2)核糖体发生和组装的精确调控在造血干细胞增殖、分化过程中的功能。

3)纳米抗体介导的骨髓增殖性肿瘤和白血病治疗。
