自我介绍
我来自英国,在英国东安格利亚大学获得博士学位,我来到亚洲从事研究工作已经有十多年了。我先是去到新加坡基因组研究所(Genome Institute of Singapore),研究胚胎干细胞及其在再生医学中应用。然后搬到日本,在大阪大学的免疫学前沿研究中心(Immunology Frontier Research Centre)主要研究免疫系统。2013年来到中国,于中国科学院广州生物医药与健康研究院(GIBH),我重新回到了胚胎干细胞的研究领域。之后来到了深圳,在南方科技大学(SUSTech)任职助理教授,同时从事研究和教学工作。
我的研究结合计算机技术和生物实验技术,全局性地研究和理解组织不同的细胞类型,了解细胞在正常状态下的相互作用和细胞在患病时如何发生错误。我特别感兴趣的方向分别有内源性逆转录病毒,对基因表达的复杂性进行建模和使用单细胞基因组学来理解细胞类型。

 

研究领域
我的实验室目前有两个目标:
第一目标,我们将创建复杂人类疾病的模型,如克罗恩氏病和帕金森病。这些疾病不是简单性遗传的,似乎是来源于DNA,环境和细胞间相互作用的高度复杂混乱的组合。我们不明白是什么原因导致这些疾病,甚至对这种类的疾病只有一个不太可靠的了解。在实验室中,我们尝试使用基因组编辑技术和计算机技术来帮助我们理解是什么导致这些疾病。
第二目标是理解为什么我们有那么多“无用的”DNA。我们基因组的编码基因只占大约4%,另外96%似乎并不重要,这其中包括大量旧病毒复制插入基因组的遗留片段,和其他寄生虫DNA片段。我(和其他人),认为所有这些额外的DNA实际上是非常重要的,而我们正试图找出它们是怎么回事和为什么是这样。
这两个部分似乎完全无关,但实际上他们是相同的。许多DNA突变与复杂疾病相关,如克罗恩氏病、帕金森病和阿尔茨海默氏症。很多DNA突变并不在是基因里面的,相反,这些突变发生在96%的“无用的”DNA序列中。我们已经开始了解所有这些96%“无用的”DNA为何而存在,也许通过理解这一点,我们也会更加了解复杂疾病。
